Abstract
Plants of Scutellaria genus have traditionally been used in the context of traditional medicine due to their pharmaceutical properties. The main active components with pharmaceutical effects found in the roots of Scutellaria plants belong to the flavonoid category, with baicalin and baicalein being the most prominent. The subject of this thesis is related to the extraction and analysis of medicinal substances from the roots of Scutellaria plants using different solvents. For this purpose, roots from Scutellaria rupestris, S. albida and S. galericulata, collected from the wild were used as well as roots form, S. rupestris plants grown in vitro. The root samples, which were initially dried and ground, were extracted using three different solvents: methanol, ethanol-water (70:30) and acetone. The plant extracts were analyzed using spectrophotometric methods to determine total phenolic and flavonoid content and antioxidant activity. Additionally, a quantitative analysis of the main pharmaceutical compounds in the methanolic extracts of Scutellaria roots was performed using high performance liquid chromatography (HPLC). The results of the spectrophotometric analyses showed that methanol was the best solvent for extracting pharmaceutical compounds, with S. rupestris (in vitro) samples exhibiting the highest concentrations of total phenolics and flavonoids. However, the antioxidant activity analysis indicated that all extracts had similar IC50 values, regardless of the extraction solvent. The analysis of pharmaceutical compounds in the extracts using HPLC revealed that, Baicalin concentrations were higher than those of baicalein in all methanolic extracts, with the S. albida strain showing the highest concentration of baicalin and simultaneously the lowest concentration of baicalein.
